Screening of a genome-reduced Corynebacterium glutamicum strain library for improved heterologous cutinase secretion

  • The construction of microbial platform organisms by means of genome reduction is an ongoing topic in biotechnology. In this study, we investigated whether the deletion of single or multiple gene clusters has a positive effect on the secretion of cutinase from Fusarium solani pisi in the industrial workhorse Corynebacterium glutamicum.
  • A total of 22 genome-reduced strain variants were compared applying two Sec signal peptides from Bacillus subtilis. High-throughput phenotyping using robotics-integrated microbioreactor technology with automated harvesting revealed distinct cutinase secretion performance for a specific combination of signal peptide and genomic deletions.
  • The biomass-specific cutinase yield for strain GRS41_51_NprE was increased by ~ 200%, although the growth rate was reduced by ~ 60%. Importantly, the causative deletions of genomic clusters cg2801-cg2828 and rrnC-cg3298 could not have been inferred a priori. Strikingly, bioreactor fed-batch cultivations at controlled growth rates resulted in a complete reversal of the screening results, with the cutinase yield for strain GRS41_51_NprE dropping by ~ 25% compared to the reference strain.
  • Thus, the choice of bioprocess conditions may turn a ‘high-performance’ strain from batch screening into a ‘low-performance’ strain in fed-batch cultivation. In conclusion, future studies are needed in order to understand metabolic adaptations of C. glutamicum to both genomic deletions and different bioprocess conditions.

Genetic analysis of persistency indices of milk yield in Jersey crossbred cattle

The study reported in this Research Communication was conducted to estimate genetic parameters for different lactation persistency indices and their association with reproductive traits in Jersey crossbred cattle. Data on lactation traits (part lactation yields and 305-days milk yield) comprising all lactations as well as reproductive traits viz. calving interval, conception rate and gestation length of 378 Jersey crossbred cattle, maintained at National Dairy Research Institute, Kalyani, West Bengal, India, were collected over a period of 35 years (1982-2016).
Five lactation persistency indices of animals (P21, P31, P32, P4 and P5) were calculated using ratio method. A total of six different animal models, ignoring or including maternal genetic or permanent environmental effects, were fitted for all persistency traits. The best model was chosen after testing the improvement of the log-likelihood values.
The estimates of direct heritability were low in magnitude and ranged from 0.02 to 0.14 for the studied traits under the best fitted animal model. The permanent maternal environmental (c2) effects of different lactation persistency indices accounted for 2-9% of the total phenotypic variance in this study. Estimated genetic correlations of lactation persistency indices with all studied reproductive traits were low to moderate and negative (-0.11 to -0.68), except for calving interval.
Phenotypic correlations of lactation persistency measures with studied reproductive traits were low (0.01 to 0.15). The low heritability estimates of all the persistency indices indicate some limited scope of genetic improvement of lactation persistency of animals through selection under the prevailing management conditions.
Genome Wide Analysis of the Transcriptional Profiles in Different Regions of the Developing Rice Grains
Background: Rice is an important food source for humans worldwide. Because of its nutritional and agricultural significance, a number of studies addressed various aspects of rice grain development and grain filling. Nevertheless, the molecular processes underlying grain filling and development, and in particular the contributions of different grain tissues to these processes, are not understood.
Main text: Using RNA-sequencing, we profiled gene expression activity in grain tissues comprised of cross cells (CC), the nucellar epidermis (NE), ovular vascular trace (OVT), endosperm (EN) and the aleurone layer (AL). These tissues were dissected using laser capture microdissection (LCM) at three distinct grain development stages. The mRNA expression datasets offer comprehensive and new insights into the gene expression patterns in different rice grain tissues and their contributions to grain development.
Comparative analysis of the different tissues revealed their similar and/or unique functions, as well as the spatio-temporal regulation of common and tissue-specific genes. The expression patterns of genes encoding hormones and transporters indicate an important role of the OVT tissue in metabolite transport during grain development. Gene co-expression network prediction on OVT-specific genes identified several distinct and common development-specific transcription factors. Further analysis of enriched DNA sequence motifs proximal to OVT-specific genes revealed known and novel DNA sequence motifs relevant to rice grain development.
Conclusion: Together, the dataset of gene expression in rice grain tissues is a novel and useful resource for further work to dissect the molecular and metabolic processes during rice grain development.

Description: CSK is a cytoplasmic tyrosine kinase that has been shown to downregulate the tyrosine kinase activity of the c-src through tyrosine phosphorylation of the c-src carboxy terminus . A yeast 2-hybrid system has been used to identify proteins associated with CSK.
